TcellimmunoreceptorwithIgandITIMdomains(TIGIT),alsocalledVSIG9andVstm3,isamemberoftheCD28familywithintheIgsuperfamilyofproteins.TIGITcontainsanimmunoglobulinvariabledomain,atransmembranedomainandanimmunoreceptortyrosine-basedinhibitorymotif(ITIM),andisexpressedonregulatory,memory,activatedTcellsandNKcells.TIGITbindstoCD155(PVR)thatappearondendriticcells(DC),macrophagesandendotheliumwithhighaffinity,andCD112(PVRL2)withloweraffinity,butnotCD113(PVRL3).TIGIT-FcfusionproteincouldinteractwithPVRonDCandenhancethesecretionofIL-10,butinhibitthemacrophageactivation.MicelackingTIGITshowincreasedTcellresponsesandsusceptibilitytoautoimmunechallenges,whileknockdownofTIGITwithsiRNAinhumanmemoryTcellsdidnotaffectTcellresponses.
