Protocol

1.SamplePreparation

Blood,Serum,Plasma,otherbodyfluid,orcellculturesupernatantcanbemeasureddirectlybyaseriesofdilutionsofthesample(1/2;1/4;1/8;…)toensurethereADIngsarewithinthestandardcurverange.YoursamplescanbedilutedwithdH2OordilutedPyruvateAssayBuffer.However,keepinmindifsamples(likehemolyzedserum/plasma)containhighleveloflactatedehydrogenasecapableofconvertingpyruvatetolactate,itisimportantforthesamplestobedeproteinated.

Add50μlofsamplestoeachwell.Wesuggestthatsamplesbeassayedinduplicate.

Note:
Ifpreparedsamplesarenotassayedthesameday,storethesamplesat-80?C.Ifsamplesneedtobedeproteinated,makesuretodeproteinizethesamplespriortostoringinthefreezer.Thedeproteinatedsampleswillbestableforonemonthwhilestoredat-80?C.Forfrozensamples,dilutionsofsamplesmustbedonerightbeforeassaying.

2.StandardPrepation

Add50µl,40µl,30µl,20µl,10µl,5µl,1µl,and0µlofdilutedPyruvateStandardtoeachwell.Thenadjustvolumeto50μl/wellwithdilutedPyruvateAssayBuffer.

WesuggestthatPyruvateStandardsbeassayedinduplicate.Astandardcurvehastobeincludedwithsamplesineachassay.

3.ReactionSolutionPreparation

50μlReactionSolutionisrequiredforeachwell,and1mlofReactionSolutionispreparedwiththefollowings:

b.MakeastandardcurvebyplottingOD570nmvaluesfromeachPyruvateStandardsasafunctionofPyruvateconcentration.Thiscanbedonewithexcelspreadsheet.

c.CalculatethevalueofPyruvateinsamplesusingtheequationobtainedfromthelinearregressionofthestandardcurve.
